Sorry for the long thread name...
I have linux Mint 19.1 FULL install to a USB. Is there any way to create a partition on my main hard drive, that can be used by linux? Possibly some of the folders for newly added programs? To increase available space beyond the USB limit?

G'day @JTByTheSea and welcome to linux.org :)

You won't get anything like the full performance on a USB stick that you could get with an install to HDD, but yes, you can.

You can use GParted on your USB to create a partition on your HDD, format it to EXT4, and then place Linux data there.
