If I install Linux Mint on my computer, not a pen-drive, will I still have to create persistence storage to save my work, file/folders/etc.

The answer is no, if you install Linux to your machine as a duel/multi boot you choose which OS boots, each will have its own space on the hard-drive[ you can partition it yourself or let the mint installer do it for you, Linux will at your command be able to access files and folders [but not programs] on your Microsoft system, but Windows will never see your Linux files.

A backup is always a good idea, but if you are comfortable with saving and finding your files on Mint you shouldn't have problems unless you don't allow Mint to shut down or reboot properly or because it locks up and you must reboot it with the off switch or by disconnecting its power before the system can save the file. What you type may be almost completely saved by the application in a backup, but some just waits until you tell it to put it in a file somehere. If it is important a separate backup is a excellent investment of you time and money. Just make sure the file system is something both linux and windows can read, e.g. FAT FAT32, ntfs, Windows 8 or later.
 
Mint is not Tails. You will need to create a home directory for every user on your system. Then each user will have their own space for their files. See the man page for adduser(8).
